AbstractThe purpose of this experiment is to use the MTT method to detect therelationship between the mycelium of the mushroom medium and the activity ofsuccinate dehydrogenase (SDH),so as to introduce the mycelial biomass of themushroom medium at different pH levels.In the experiment,5188 strains of slidingmushrooms were used as experimental materials.First,the suitable mediumformulation and mycelium cultivation method were found through screening.Secondly,the liquid cultured sliding mushroom mycelium balls were weighed 0.05g,0.1g,0.15g,0.2 respectively.g,0.25g,0.3g,0.35g,0.4g using the MTT method todetect the standard curve between succinate dehydrogenase (SDH)and the mushroomhyphae,with lime (0%,1%,2%,3 %4%,5%,6%)to adjust the pH of the solidmedium,and then use this curve as a standard to obtain whether the mycelial biomassof the mycelia of the solid medium of the pleurotus ostreatus has changed underdifferent acid and alkali conditions.The experimental results show that:in the rangeof 1%-3%of lime,the mycelial biomass generally shows an upward trend,reachingthe highest when the amount of lime is 3%.When the amount of lime reached 4%andabove,the mycelial biomass decreased.Therefore,the medium with a lime content of3%is most suitable for mycelial growth.This experiment is to provide a theoreticalreference for the MTT method to determine the biomass of edible fungus mycelium.Key words:Pholiota nameko:MTT:Mycelial biomass;Succinate dehydrogenase
