北京理工大学珠海学院2020届本科生毕业设计Study on rapid detection method ofantiviral drug residues in eggsAbstractIn the experiment,the main components of eggs and the structural properties of antiviraldrugs were discussed.In other matrix test experience,a more rapid and reasonable extractionand purification step was obtained.In combination with liquid chromatography-tandem massspectrometry (LC-MS/MS)instrument analysis,the selection of liquid chromatographycolumn,gradient elution solution and sequence parameters,mass spectrometry conditions,etc.were optimized to determine the residual gold in eggs.Amantadine and amantadine werequantitatively analyzed and detected by internal standard method to achieve rapid detectiontechnology.The extraction method was determined as follows:adding 15 mL oftrichloroacetic acid-acetonitrile (9:1)mixed extract to 5 g egg sample,ultrasonication for 10min,whirlpool for 3 min,and centrifugation for 5 min at 10,000 r/min in a high-speedcentrifuge at 20 C.The purification steps are as follows:5 mL of 2%formic acid and 3 mL of1%acetonitrile formic acid are sequentially added into the activated PCX column,the PCXcolumn is vacuum-dried,eluted with 4 mL ammonia-ammonium acetate-methanol mixedsolution,dried at 50 C by a nitrogen concentrator,0.1%formic acid-methanol and water areadded into the 1 mL solution,and mixed by vortex for 30 s and then passed through 0.22 mmembrane.The experimental results showed that the average recoveries of the two antiviral drugswere more than 80%,the recoveries of amantadine were as high as 85%,RSD was0.118-0.187,and the recoveries of amantadine were 81%-98%and RSD was 0.038-0.134under the gradient of 1,2,5 and 10 ug/kg.The standard curves of the two antiviral drugsshowed a good linear relationship in the range of 0-20 ug/kg,with R20.999.Keywords:LC-MS/MS;Amantadine;Rimantadine;egg
