AbstractIgM is the main immunoglobulin in large yellow croaker,which plays a vital rolein the immune system of fish.In this thesis,the serum IgM of Larimichthys croceapurified by protein A affinity,according to the standard immune process,was used asantigen to immunize Balb/c mice,and the antiserum,polyclonal antibody,wasacquired.The valence of mouse antiserum determined by enzyme-linkedimmunosorbent assay (ELISA)achieved 1:20000.Western blotting was used toanalyze the binding characteristics of murine polyclonal antibodies against serum IgMof L.crocea.The results showed that the polyclonal antibodies could specificallycombine with heavy and light chains of IgM L.crocea,which indicated that itprepared in this experiment had good specificity and could be applied to subsequentexperimental studies.The preparation of rat polyclonal antibody against the serumIgM of L.crocea helps to understanding of the immune system of the fish,and willalso lay a foundation for the study of the immune mechanism and related immunedetection of L.crocea.Key words:Larimichthys crocea;IgM;mice polyclonal antibody;ELISA;Westernblotting
