Identification of Elephant Species by Nested PCR and GeneSequencingAbstractIn order to establish a technical method that can accurately identify elephant species,thisstudy designed and screened two pairs of specific primers for the conserved sequences ofelephant species,and established nested PCR combined sequencing for elephant species.Identification method.And by using different samples and samples of different concentrationsto test the method for specificity and sensitivity,and finally using the system and procedureestablished in this experiment,the three ivory samples of the technical center were testedthree times repeatedly.After specificity and sensitivity tests and clinical sample testing,thefollowing relevant results were obtained.The specific results show that the method can onlydetect DNA samples of modern elephant and mammoth ivory,and does not amplify DNAsamples of other animal species with similar teeth and bone structures;The sensitivity testresults show that the detection sensitivity of the nested PCR target animal species DNAsamples is 10 times higher than that of fluorescent PCR;Three clinical samples can get bettersequencing results,which can clearly identify the species of modern elephants and mammoths.The nested PCR method established in this study can effectively and accurately identify ivoryand ivory products,which is helpful for the supervision of domestic trade in ivory and ivoryproducts.When there are ivory and its products in circulation,it can be identified according tothis method.Keywords:Gene sequencing;Quantitative Real-time PCR;Elephantidae
