Researchon TMPyP4inducingCellsAutophagyAuthor:GuoManfeiTutor:Huang Weiwei(NorthwestA&FUniversity,Yangling,Shaanxi,712100)Abstract:Objective:to explore the relationship between TMPyP4 andautophagy of tumor cells andthe signaling pathway affected by its induction of autophagy.Methods:the effects of TMPyP4 on theexpression of autophagy related proteins and the formation of autophagosomes in tumor cells and thesignaling pathways were detected by westernblotting and fluorescence observation.Results:the tumor cellsto detect autophagy markerprotein LC3IIB level rise obviously,and autophagic degradationprotein p62level significantly decrease,and cytolysosome and autophagy-lysosome increase by the fluorescencedetectionTMPyP4 can induce autophagy flow in tumor cells through the combination of autophagyupstream inhibitor and downstream inhibitor with TMPyP4.By detecting the upstream mTOR signalingpathway of TMPyP4,it was found that the level of p-ampka T172 significantly up-regulates,the content ofAMPK adecreases,and the expression of p-mtor S2448and mTOR down-regulates.Conclusion:the aboveresults suggest that TMPyP4 can induce autophagy of tumor cells and generate autophagy flow.TMPyP4inhibits mTOR induction of autophagy in tumor cells by activating the AMPK signaling pathway.Keywords:Autophagy:TMPyP4:Hep-2细胞：AMPK
