AbstractObjective:Cardiovascular disease is one of the leading causes of death in the world and poses aserious threat to human health.Acute myocardial infarction is one of the most seriouscardiovascular disease.It has been found that stromal cell derived factor 1 might increase postmyocardial infarction to recruit bone marrow stem cell to repair and regeneration of damagedtissues.The aim of this study was to explore the changes of SDF-1 expression levels with the timeof myocardial infarction and the expression of SDF-1 in different focal areas.Methods:Themyocardial infarction model of SD rats was established by ligation of left descending coronaryartery,and the rat heart was harvested after 0.5 days,1 days,2 days,3 days,5 days and 7 days,respectively.Real-time fluorescence quantitative PCR(RT-gPCR),western blot(WB)andenzyme-linked immunosorbent assay(ELISA)were used to detect the expression of SDF-1 gene andprotein in the infarct area,the junctional area and the normal area of the heart at different timepoints after myocardial infarction.Results:The expression levels of SDF-1 gene and protein in theinfarct area and junctional area increased gradually with time,and reached the peak level on day 2.The expression level of SDF-1 was the most obvious in infarct area,but not in normal area.Conclusion:Post of myocardial infarction,the expression of SDF-1 gene and protein increased firstand then decreased in the infarct area and the junctional area.Keywords:Myocardial infarction;Stromal cell derived factor 1;Real-time fluorescencequantitative PCR;Western blot;Enzyme linked immunosorbent assay.
