高效液相色谱法测定湘产黄精中薯蓣皂苷元含量研究目录摘要.4引言…。.61实验材料和方法.61.1实验材料.61.1.1实验样品.…61.1.2实验试剂.61.1.3实验仪器..612实验方法..,91.2.1样品处理....91.2.2色谱条件.…..91.2.3供试品溶液的配制…101.2.4对照品溶液的配制.102薯蓣皂苷元含量的测定.…102.1线性实验.….102.2精密度实验.102.3重现性实验…102.4稳定性实验.102.5加样回收实验102.6建立指纹图谱102.7样品含量测定.….113实验结果…113.1线性实验结果.113.2精密度实验结果.….113.3重现性实验结果.113.4稳定性实验结果.113.5加样回收率实验结果.…113.6不同湘产黄精指纹图谱的建立.…123.7薯蓣皂苷元含量测定的结果183.7不同品种黄精的质量差异分析..204分析与总结.……214.1样品前处理条件及检测波长的选择.214.2色谱柱的选择.……214.3流动相的选择…214.4分析与小结…22参考文献..24致谢.…251摘要摘要目的基于HPLC指纹图谱技术和统计学分析方法,建立指纹图谱并进行相似度评价,测定来源不同的湘产黄精中薯蓣皂苷元的含量。方法使用SB-C18 AnalyticalHPLC Column色谱柱(250mm×4.6mm,5m),选择202nm为检测波长、乙腈(A)-0.1%磷酸水溶液(B)为流动相,进行梯度洗脱,体积流量1.0 mL/min,柱温为30℃:测定不同来源湘产黄精药材薯蓣皂苷元含量,建立不同品种湘产黄精HPLC指纹图谱并进行比较分析,同时针对不同品种黄精的薯蓣皂苷元含量进行显著性差异分析。结果采用HPLC-DAD建立了不同来源黄精药材指纹图谱并对薯蓣皂苷元进行含量测定,通过比对分析发现不同来源品种间相似度,多花黄精与卷叶黄精相似度最低为0.880,与湘黄精、长梗黄精的相似度分别为0.984、0.981;发现湘黄精与多花黄精相比相似度最高;湘黄精与卷叶黄精的相似度为0.787,与长梗黄精相似度为0.940;湖南省安化市樟楠湾所产的多花黄精的薯蓣皂苷元的含量最高;基于薯蓣皂苷元含量指标分析表明:湘黄精组与卷叶黄精、多花黄精、长梗黄精组不存在差异。结论研究结果显著表明,湘产黄精中薯蓣皂苷元含量因产地、品种而异,其中多花黄精含量居首,该研究所用方法可行,可为黄精药材的质量控制和品质评价工作提供有力的技术支撑;基于薯蓣皂苷元含量差异分析,湘黄精与其他三个品种均不存在差异,为湘黄精的化学成分与黄精药材来源种相似,可作为药材代用品提供科学依据。关键词黄精;HPLC;指纹图谱;薯蓣皂苷元AbstractObjective Based on HPLC fingerprint technique and statistical analysis method,the fingerprintwas established and the similarity evaluation was conducted to determine the content of potatosaponin from different sources.Method Using SB-C18 Analytical HPLC Column column (250mm 4.6 mm,5 um),selecting 202 nm for detection wavelength and acetonitrile (A)-0.1%aqueous phosphate acid solution (B)as mobile phase,gradient elution,volume flow was 1.0 mL/min,and column temperature was 30C;Content of components of HPLC fingerprints ofdifferent varieties were established and comparative analysis of different varieties.ResultsHPLC-DAD was used to establish the fingerprint map of different sources and determine the2content of potato saponin element,Through alignment analysis,The lowest similarity ofpolyflower and yellow was 0.880,The similarity with Hunan and long stem yellow were 0.984and 0.981 respectively;It is found that xiang yellow essence has the highest similaritycompared with Hua flower yellow essence;The similarity was 0.787,The similarity with thelong stem yellow essence was 0.940;Hunan province has the highest content of potato saponin;Based on the analysis of potato saponin content index,there is no difference between xiangyellow sperm group and rolling leaf yellow sperm,more flower yellow sperm and long stemyellow sperm group.Conclusion The results show that the content of potato saponins variesfrom origin and variety,with the method is feasible,which can provide strong technical supportfor the quality control and evaluation of the content of the essence,there is no differencebetween the chemical composition of the same,and can be used as a substitute for medicinalmaterials.Key words yellow essence;HPLC;fingerprint;potato saponin element3
