白术活性成分与内生微生物相关性研究摘要目的通过对不同产地白术药效活性成分、内生物多样性分析及其之间的相关性分析,探讨内生微生物与白术道地性关系,为揭示白术道地性提供依据方法利用UHPLC-MSMS技术测定白术内酯类成分、核苷氨基酸类成分含量,利用苯酚浓硫酸法测定多糖含量,利用考马斯亮蓝法测定蛋白质含量,利用高通量测序技术检测内生微生物多样性,并对成分和微生物进行相关性分析结果不同产地白术化学成分含量存在差异,其中安徽产白术(安徽种源)三种内酯类成分含量最高。安徽产白术(安徽种源)酸性多糖含量最高,但浙江产白术(浙江种源)中性多糖含量最高。浙江产白术(安徽种源)核苷类和氨基酸类成分、蛋白质含量最高。P℃oA分析结果表明不同种源及产地间白术内生细菌组成无显著性差异,白术内生真菌组成存在差异。内生真菌Cadophora、Acidomelania、Helotiales fam_Incertae sedis属丰度较高,内生细菌Burkholderia-Caballeronia-Paraburkholderia、Alcaligenaceae属丰度较高。对内生微生物和白术活性成分进行相关性分析,内生真菌Sclerotiniaceae,Atractiellales,Rhexocercosporidium和内生细菌Bosea、Xanthobacteraceae、yphomicrobium等与白术内酯类成分成正相关结论本研究丰富白术内生微生物资源,为白术内生微生物资源的挖掘提供参考。探讨内生微生物与白术活性成分的相关性,为内生菌应用于优质栽培提供数据支撑。关键词:白术,白术内酯,内生微生物,多样性分析Study on the correlation between active components ofAtractylodes macrocephala and endophytic microorganismsAbstract Objective The relationship between endophytic microorganisms andgenuine locality of Atractylodes macrocephala was studied by analyzing thepharmacodynamic active components,the diversity of endophytic microorganismsand the correlation among them,so as to provide the basis for revealing genuinelocality of A.macrocephala.Methods UHPLC-MS/MS was used to determine thecontents of lactones and amino acids in A.macrocephala,the content ofpolysaccharides was determined by phenol concentrated sulfuric acid method,and thecontent of protein was determined by Coomassie Brilliant Blue method,theendophytic microbial diversity was detected by high-throughput sequencing and itscorrelation was analyzed.Results The three atractylenolide contents of A.macrocephala from Anhui were the highest.The acid polysaccharidecontent of thesamples from Anhui (the seedlings came from Anhui)was the highest,and the neutralpolysaccharide content of the samples from Zhejiang (the seedlings came fromZhejiang)was the highest.The total content of nucleosides,amino acids and proteinin A.macrocephal from Zhejiang (the seedlings came from Anhui)was the highest.The results of PCOA analysis showed that the composition of endophytic bacteria inA.macrocephala from different provenances and habitats had no significantdifference,but the composition of endophytic fungi had significant difference,and theabundance of Cadophora,Acidomelania,Helotiales in endophytic fungi was higher,endophytic bacteria Burkholderia-Caballeronia-Paraburkholderia,Alcaligenaceaeare abundant.Component correlation analysis based on high-throughput sequencingresults,among them,fungi Sclerotiniaceae,Atractiellales,Rhexocercosporidium,andbacteria Bosea,Xanthobacteraceae,Hyphomicrobium,were significantly andpositively correlated with the active components.Conclusion This study enriches theendogenous microbial resources of A.macrocephala,discusses the correlationbetween endophytic microorganisms and the active ingredients of A.macrocephala,and provides a reference basis for the mining of endogenous microbial resources of 4.macrocephala.Key words:Atractylodes macrocephala,atractylenolide,endophyte,diversityanalysis引言白术为菊科植物
